The LE and SC4Tool are two completely different programs. Generally both are used because they supplement each other.

-The Lot Editor is used by many to physically make the lots. This is 'lotting' all the textures, props, trees, and buildings on what you want your finished lot to look like.

-SC4Tool, on the other hand, is for modding. This has tons of uses. Some features are texture maker, Transit-Enabler (To TE your lots), Dependency checker (to show what prop packs are used in your new lot so you can document this if the lot is publicly released), and many other essential modding tools.

I would strongly recommend both. I would also highly recommend the PIM-X Tool, which contains many other tools for modding your lot. I also believe it contains a lot editor, but I haven't really tried it yet... it can be downloaded at http://sc4devotion.com/csxlex/lex_filedesc.php?lotGET=2260

hope this explanation helps!
